The novel P II -interactor PirC identifies phosphoglycerate mutase as key control point of carbon storage metabolism in cyanobacteria

چکیده
Significance In this work, we identified the regulatory mechanism of key control point cyanobacterial carbon metabolism, glycolytic phosphoglycerate mutase (PGAM) reaction, converting 3-PGA into 2-PGA and thereby exporting organic from photosynthetic Calvin cycle. We show that PGAM activity is controlled by a small modulator protein PirC (product sll0944 ), which inhibits enzyme through protein–protein interaction. The availability for inhibition pervasive carbon/nitrogen balance regulator P II , sequesters at low 2-oxoglutarate levels releases it high levels. PirC-mediated triggers glycogen accumulation, disrupting regulation allows redirection flux, decisive requirement transforming cyanobacteria green factories.

متن کاملDiscovery and analysis of cofactor-dependent phosphoglycerate mutase homologs as novel phosphoserine phosphatases in Hydrogenobacter thermophilus.
Phosphoserine phosphatase (PSP) catalyzes the dephosphorylation of phosphoserine to serine and inorganic phosphate. PSPs, which have been found in all three domains of life, belong to the haloacid dehalogenase-like hydrolase superfamily. However, certain organisms, particularly bacteria, lack a classical PSP gene, although they appear to possess a functional phosphoserine synthetic pathway. The...
متن کاملThe catalytic bimodality of mammalian phosphoglycerate mutase.
Rabbit muscle phosphoglycerate mutase, presumed to manifest an absolute cofactor requirement for activity, has been found to express catalysis (3 +/- 1% of optimum) in the absence of added D-glycerate-2,3-P2. Isotope experiments indicate that this catalysis proceeds through a binary phosphoryl enzyme-glycerate intermediate which dissociates into free enzyme and monophosphoglycerate. 32P-Labeled...

BACKGROUND Phosphoglycerate mutase (PGAM) deficiency (glycogen storage disease type X) has been reported in 12 patients of whom 9 were African American. OBJECTIVE To describe 2 patients, 1 of Pakistani and 1 of Italian ethnic origin, with typical clinical and biochemical changes of glycogen storage disease type X and novel mutations in the gene encoding the muscle subunit of PGAM (PGAM2). D...
متن کاملThe role of the C-terminal region in phosphoglycerate mutase.
Removal of the C-terminal seven residues from phosphoglycerate mutase from Saccharomyces cerevisiae by limited proteolysis is associated with loss of mutase activity, but no change in phosphatase activity. The presence of the cofactor 2, 3-bisphosphoglycerate, or of the cofactor and substrate 3-phosphoglycerate together, confers protection against proteolysis. The substrate alone offers no prot...
